Ans A) When a bacteria affect or infect a cell then the defence mechanism of the that infected cell tries to protect itself from the bacteria by fusing the lysosome to form phagolysosome. The lysosome contain various enzyme can protect the infected cell by destroying the the bacteria. Lysozyme is one of the enzyme present in the lysosome that has ability to destroy the cell by breaking the cell wall. The walls of Gram- Positive bacteria can be easily destroyed when treated with lysozymes which selectively breaks the peptidolycan cell wall. But the gram- negative bacteria are resistance to lysozyme activity due to the presence of an outer membrane that surround the thin peptidoglycan membrane. This outer membrane is composed re rich lipids and large protein molecules cannot penetrate the outer membrane. Thus the gram- nagative bacteria has a resistance to phagocytosis untill its outer lipid rich membrane is intact.

Ans B) Bacterial cell wall with regard to chemical composition is:-

The bacterial cell wall is a rigid stucture with gives shape to the cell. The cell wall is mainly composed of peptidoglycan which is also termed as \"murein\". They are the insoluble, porous and cross linking polymer of carbohydrate derivatives, amino acids. The main component of cell wall is N-acetylglucosamine (NAG), N-acetylmuramic acid (NAM), alanine, glutamate. The cross linking between these component form the tough and rigid structure of bacterial cell wall. Accoeding to chemical composition they are termed as Gram- Positive or Gram Negative bateria. Gram -positive bacteria has cross linking NAG,NAM and other amino acid to form the complex cell wall structure. Gram- Negative bacteria are rich in lipids, lipopolysaccharides and an outer membrane that surrounds a thin underlyng layer of peptidoglycan.

Ans C) Addition of alcohol, a decolourizer to the gram postive bacteria resulting in the retention of crystal violet-iodine complex by closing the pore of the cell wall. In gram- negative bacteria contain higher lipid content and the alcohol dissolve the lipid content. The primary stain is able to leach into the solvent and alcohol wash away the dye and leaving the gram-negative bacteria unstrained.
